James Johnstone, 5th of Elphinstone was born before 1531.2 He was the son of Andrew Johnstone, 4th of Elphinstone and Margaret Douglas.2 He married Margaret Ruthven, daughter of William Ruthven, 2nd Lord Ruthven and Janet Halyburton, Lady Dirletoun, after 24 March 1550.1,2 He died between 1570 and 1575.2
