Sir John Cotton, 2nd Bt. was born in 1647. He was the son of Sir John Cotton, 1st Bt. and Jane Hinde.1 He married Elizabeth Sheldon, daughter of Sir Joseph Sheldon and Elizabeth Clifton, on 14 January 1678/79 at Westminster Abbey, Westminster, London, EnglandG.1 He died on 20 January 1712/13 at Landwade, Cambridgeshire, EnglandG.1 He was buried on 23 January 1712/13 at Landwade, Cambridgeshire, EnglandG.1 He died intestate and his estate was administered on 24 April 1716.1
He succeeded as the 2nd Baronet Cotton, of Landwade, co. Cambridge [E., 1641] in 1689.1 He held the office of Member of Parliament (M.P.) for Cambridge from 1689 to 1708.1 He held the office of Recorder of Cambridge.1

He graduated from Cambridge University, Cambridge, Cambridgeshire, EnglandG, in 1705 with a Master of Arts (M.A.)1 He held the office of Member of Parliament (M.P.) for Cambridge between 1708 and 1722.1 He was a Lord Commissioner of trade and plantations in 1712.1 He succeeded as the 3rd Baronet Cotton, of Landwade, co. Cambridge [E., 1641] on 20 January 1712/13.1 He held the office of Member of Parliament (M.P.) for Cambridgeshire between 1722 and 1727.1 He held the office of Member of Parliament (M.P.) for Cambridge between 1727 and 1741.1 He held the office of Treasurer of the Chamber in 1744.1 He held the office of Member of Parliament (M.P.) for Marlborough between 1747 and 1752.1
